阿尔茨海默病（Alzheimer disease，AD）以病理学以特征性的老年斑和神经原纤维缠结为主，故β淀粉样蛋白（Aβ）和过度磷酸化的tau蛋白（tau）作为AD的主要标志。然而近年来在许多尸检结果中发现，许多有认知障碍的老年人脑组织中，只有神经纤维缠结（NFT）单独存在，没有或仅有少许淀粉样蛋白（Aβ）斑块，这种“NFT+/Aβ-”模式不符合AD神经病理改变的正式标准。
